    检验原理

    Test principle

    该培养基含有乳糖、蔗糖和葡萄糖,只能利用葡萄糖的细菌,葡萄糖被分解产酸使斜面先变黄,但因量少,生产的少量酸。因接触空气而氧化,加之细菌生长利用培养基中含氮物质,生成碱性产物,故斜面后来又变红。底部由于厌氧状态,酸类不被氧化,仍保持黄色。而发酵乳糖的细菌,则产生大量的酸,使整个培养基呈黄色。另外,因某些细菌能分解含硫氨基酸,生成硫化氢,硫化氢和培养基中的铁盐反应生成黑色的硫化亚铁沉淀。

    The medium contains lactose, sucrose and glucose, which can only be used by bacteria. The glucose is decomposed to produce acid, which turns the slope yellow first, but because of the small amount, a small amount of acid is produced. Due to the oxidation of contact with air, coupled with the growth of bacteria using nitrogenous substances in the medium to produce alkaline products, so the slope later turned red. At the bottom, due to the anaerobic state, the acids are not oxidized and remain yellow. Bacteria that ferment lactose, on the other hand, produce a lot of acid, making the whole medium yellow. In addition, because some bacteria can decompose sulfur-containing amino acids to produce hydrogen sulfide, hydrogen sulfide and the iron salt in the medium reaction to produce black ferrous sulfide precipitate.

    使用方法

    Usage

    称取本品63.4g,加入蒸馏水1000ml中,搅拌加热煮沸至完全溶解,分装,121℃高压灭菌15分钟制成高低层2-3厘米的斜面备用。(中国药典)

    Weigh 63.4 g of the product, add to 1000 ml of distilled water, and stir while heating to boiling until completely dissolved. Dispense into tubes to form high or low slants (2-3 cm), and sterilize in an autoclave at 121°C for 15 minutes. (Chinese Pharmacopoeia)
